The identification of a sugar derived from coenzyme B12.

چکیده
The structure of coenzyme Biz has been shown to be based upon that of vitamin Blz (1). The main structural differences between the coenzyme and the vitamin are the absence of cyanide and the presence of an adenine nucleoside in the coenzyme. An adenine nucleoside was isolated after photolysis of the coenzyme (2, 21). Although the photolysis reaction is complex and gives rise to two colorless ultraviolet-absorbing compounds, the adenine nucleoside (peak 1 photolysis product) is generally the more abundant. Mild acid hydrolysis of the coenzyme yields adenine and a sugarlike compound. Adenine was identified by its ultraviolet absorption spectrum, its chromatographic and ionophoretic behavior (2), and by preparation of its picrate (3). Further elucidation of the structure of the cobamide coenzymes requires the identification of the sugar-like compound formed by acid hydrolysis. This paper describes the characterization of this compound as D-erythro-2,3-dihydroxy-A4-pentenal.

Effects of Vitamin B12 in Neonates and Young Infants
Vitamin B12 (cobalamin) is an essential coenzyme for nucleic acid synthesis. Animal protein is the major dietary source of vitamin B12. Deficiency of vitamin B12 leads to megaloblastic anemia, degeneration of the brain, spinal cord, peripheral nerves, and abnormalities of epithelial tissues. Two factors are necessary for the cure of megaloblastic anemia: one in food (extrinsic factor) and one i...

A member of the vitamin B12 group has been identified as the red chromophore of reddish brown enzyme fractions obtained from Clostridium sticklandii. An orange compound which appears to be identical with the coenzyme form of pseudovitamin B12 (or adenine-B12 coenzyme) discovered by Barker et al. (Proe. Natl. Acad. Sci. U. S., 44, 1093, 1958) is obtained if preparations are protected from visibl...
متن کاملIdentification of the human and bovine ATP:Cob(I)alamin adenosyltransferase cDNAs based on complementation of a bacterial mutant.
In humans, deficiencies in coenzyme B12-dependent methylmalonyl-CoA mutase (MCM) lead to methylmalonyl aciduria, a rare disease that is often fatal in newborns. Such deficiencies can result from inborn errors in the MCM structural gene or from mutations that impair the assimilation of dietary cobalamins into coenzyme B12 (Ado-B12), the required cofactor for MCM. ATP:cob(I)alamin adenosyltransfe...
متن کاملHematologic Effect of 5,6-dimethyl-benzimidazolyl Cobamide Coenzyme in a Patient with Pernicious Anemia.
Since 5, 6-dimethylbenzimidazolyl cobamide coenzyme (coenzyme B12) as one of the coenzyme forms of vitamin B12 was reported by Barker et al. (1) for the first time in 1960, a good deal of works have been carried out on its biochemical and physiological roles (2-8). The recent knowledge concerning the coenzyme has revealed that it is an active form of vitamin B12 (5, 6-dimethylbenzimidazolyl cob...
متن کاملPurification and Characterization of Glycerol Dehydratase from Lactobacillus reuterit
A coenzyme B12-dependent glycerol dehydratase from Lactobacillus reuteri has been purified and characterized. The dehydratase has a molecular weight of approximately 200,000, and sodium dodecyl sulfatepolyacrylamide gel electrophoresis yielded a single major band with a molecular weight of 52,000. Km values for substrates and coenzyme B12 were in the millimolar and the submicromolar range, resp...
